【樣品收集】樣品如果不立即分析，應分裝后冷凍保存，且避免反復凍融。
血清——用干凈試管收集血液，室溫凝固2小時或4℃過夜，離心1000×g 10分鐘，收集血清。立即分析或分裝后-20℃冷凍保存。
細胞培養、組織勻漿、體液——離心去除沉淀，立即分析或分裝后-20℃冷凍保存。
大鼠線粒體解偶聯蛋白1(UCP1) 加樣在ELISA中一般有3次加樣步聚，即加標本，加酶結合物，加底物。加樣時應將所加物加在LEISA板孔的底部，避免加在孔壁上部，并注意不可濺出，不可產生氣泡。加標本一般用微量加樣器，按規定的量加入板孔中。每次加標本應更換吸嘴，以免發生交叉污染，也可用一次性的定量塑料管加樣。有此測定（如間接法ELISA）需用稀釋的血清，可在試管中按規定的稀釋度稀釋后再加樣。也可在板孔中加入稀釋液，再在其中加入血清標本，然后在微型震蕩器上震蕩1分鐘以保證混和。加酶結合物應用液和底物應用液時可用定量多道加液器，使加液過程迅速完成。
