視神經細胞相關蛋白抗體,Anti-DOCK3抗體保存條件:Store at -20 °C for one year. Avoid repeated freeze/thaw cycles. The lyophilized antibody is stable at room temperature for at least one month and for greater than a year when kept at -20°C. When reconstituted in sterile pH 7.4 0.01M PBS or diluent of antibody the antibody is stable for at least two weeks at 2-4 °C.
各種HRP酶標記成抗體的效價測定程序如下(如:羊抗鼠IgG-HRP的效價測定):
1、抗原包板:15ug/ml 小鼠IgG-pH9.6·CB液 0.1ml/孔;
2、37°C包被60min或4°C過夜;
3、PBST洗板3次,5min/次;
4、加入倍比稀釋的HRP標記抗體 0.1ml/孔;
5、37°C 30min;
6、PBST洗板5min/次×3;
7、底物液0.1ml/孔 37°C 5-10min;
8、終止液:2N H2SO4 0.05ml/孔;
9、450nm 測定OD值;
10、陽性OD值比對照值大3倍,為判斷效價稀釋度。

視神經細胞相關蛋白抗體背景介紹:MOCA (modifier of cell adhesion), also known as Presenilin-binding protein (PBP) or dedicator of cytokinesis protein 3 (DOCK3), is a 2030 amino acid cytoplasmic protein belonging to the DOCK family. MOCA interacts with Presenilin proteins and has the ability to stimulate Tau phosphorylation suggesting that MOCA may be involved in Alzheimer disease. MOCA is also thought to be a guanine nucleotide exchange factor (GEF) which activates small GTPases by exchanging bound GDP for free GTP. Analysis of ectopic expression suggests that MOCA may affect the function of small GTPases involved in the regulation of Actin cytoskeleton or cell adhesion receptors. MOCA is localized to the neuropil, and sometimes in pyramidal cells, in normal brains, while in Alzheimer disease brains, MOCA is present in neurofibrillary tangles.
免疫熒光染色與免疫組化前期處理相同,因為內源性過氧化物酶對免疫熒光實驗沒有影響,同時雙氧水反而會干擾到熒光素發光,所以這步操作在免疫熒光中不需要做。
