胞漿蛋白SH3GL3抗體背景介紹：The endophilins comprise a family of three SH3 domain-containing proteins designated Endophilin I, II and III, or alternatively known as SH3P4, SH3P8 and SH3P13, respectively. These proteins associate with Amphiphysin, Synaptojanin and Dynamin and are implicated in presynaptic vesicle trafficking at nerve terminals. The expression patterns of the endophilins are consistent with their cellular functions at the neuronal synapse, as Endophilin I is ex- pressed only in the brain. Both Endophilin II and Endophilin III are detected in a variety of tissues. Endophilin I is also implicated in modulating G protein-coupled receptor signaling by functioning as an adapter protein and directing 1 adrenergic receptors to the endocytic machinery.

抗體修復方式:

修復液：0.01M 枸櫞酸（pH6.0）或者0.05M EDTA（pH8.0）

抗體修復方法：

方法1：沸水浴修復，將盛有修復液和玻片的燒杯置于沸水浴環境，保持外部沸騰狀態15min，自然冷卻至室溫；

方法2：微波修復，將盛有修復液和玻片的燒杯置于微波爐中，高火5min，停火3min，中火5min，自然冷卻至室溫。

胞漿蛋白SH3GL3抗體,Anti-SH3GL3抗體免疫組化操作步驟：

（一）、儀器設備

1、 18cm不銹鋼高壓鍋或電爐或用微波爐

2、 水浴鍋

（二）、試 劑

1、 PBS緩沖液（pH7.2―7.4）

2、 0.01mol/L檸檬酸鈉緩沖液（CB,pH6.0,1000ml）

3、 3%甲醇―H2O2溶液：用30% H2O2 和80%甲醇溶液配制

4、 風裱劑：

a. 甘油和0.5mmol/L碳酸鹽緩沖液（pH9.0–9.5）等量混合

b. 油和TBS（PBS）配制

5、 TBS/PBS pH9.0–9.5，適用于熒光纖維鏡標本；pH7.0-7.4適合光學纖維標本

（三）、操作流程

1、 脫蠟和水化：脫蠟前應將組織芯片在室溫中放置60分鐘或60℃恒溫箱中烘烤20分鐘

a. 組織芯片置于二甲苯中浸泡10分鐘，更換二甲苯后在浸泡10分鐘

b. 無水乙醇中浸泡五分鐘

c. 95%乙醇中浸泡五分鐘

d. 75%乙醇中浸泡五分鐘

2、 抗原修復：用于福爾馬林固定的石蠟包埋組織芯片
