精zi細(xì)胞相關(guān)蛋白抗體保存條件：Store at -20 °C for one year. Avoid repeated freeze/thaw cycles. The lyophilized antibody is stable at room temperature for at least one month and for greater than a year when kept at -20°C. When reconstituted in sterile pH 7.4 0.01M PBS or diluent of antibody the antibody is stable for at least two weeks at 2-4 °C.

精zi細(xì)胞相關(guān)蛋白抗體,Anti-SPERT抗體抗原修復(fù)方法：

方法1：沸水浴修復(fù)，將盛有修復(fù)液和玻片的燒杯置于沸水浴環(huán)境，保持外部沸騰狀態(tài)15min，自然冷卻至室溫。
方法2：微波修復(fù)，將盛有修復(fù)液和玻片的燒杯置于微波爐中，高火5min，停火3min，中火5min，自然冷卻至室溫。
方法3：高壓修復(fù)，修復(fù)液加入高壓鍋加熱至沸騰，放入玻片，封蓋加壓持續(xù)加熱至噴氣時(shí)開始計(jì)時(shí)修復(fù)2min，自然冷卻至室溫。
方法4：使用*修復(fù)，將*修復(fù)液加入到組織上，37℃，消化30min

Western Blotting 實(shí)驗(yàn)步驟概述：

1、 組織取材：組織塊稱重

2、 利用液氮、研缽粉碎組織塊

3、 加入RIPA緩沖液（每克組織3ml RIPA），PMSF（每克組織30μl，10 mg/ml PMSF），

利用Polytron進(jìn)一步勻漿（15,000轉(zhuǎn)/分*1分鐘）維持4℃

4、 加入PMSF（每克組織30μl，10 mg/ml PMSF），冰上孵育30分鐘

5、 移入離心管4℃，約20,000g（約15,000轉(zhuǎn)）15分鐘

6、 上清液為細(xì)胞裂解液可分裝-20℃保存

7、 進(jìn)行Bradford比色法測定蛋白質(zhì)濃度

8、 取相同質(zhì)量的細(xì)胞裂解液（體積*蛋白質(zhì)濃度），并加等體積的2×電泳加樣緩沖液

9、 沸水浴中3分鐘

10、上樣

11、電泳（濃縮膠20mA，分離膠35mA）

12、電轉(zhuǎn)膜儀轉(zhuǎn)膜（100mA 40分鐘）

13、膜用麗春紅染色，膠用考馬斯亮藍(lán)染色

14、Westernblot 試劑盒顯色

背景介紹：A variety of morphological and molecular changes are required for spermatozoa formation. These steps are temporally guided by the transcription and translation of several testis-specific genes. SPERT (spermatid associated), also known as CBY2 (chibby homolog 2), spermatid flower-like structure protein or NURIT, is a 448 amino acid novel leucine-zipper protein belonging to the chibby family of proteins.Expressed uniquely in the spermatid flower-like structure, SPERT interacts with Nek1, a member of the NIMA-family kinase family that is associated centrosomal stability and ciliogenesis. Containing a leucine-zipper motif and two coiled-coil regions, SPERT is transcribed through the elongation stage of the spermatids. SPERT is absent from mature spermatozoa and is thought to be involved in transporting proteins that are to be discarded via the residual bodies.
