儲(chǔ) 存液: 0.01M TBS(pH7.4) with 1% BSA, 0.03% Proclin300 and 50% Glycerol

保存條件: Store at -20 °C for one year. Avoid repeated freeze/thaw cycles. The lyophilized antibody is stable at room temperature for at least one month and for greater than a year when kept at -20°C. When reconstituted in sterile pH 7.4 0.01M PBS or diluent of antibody the antibody is stable for at least two weeks at 2-4 °C.

KIR2.3抗體-鉀離子通道KIR2.3抗體產(chǎn)品介紹：Several different potassium channels are known to be involved with electrical signaling in the nervous system. One class is activated by depolarization whereas a second class is not. The latter are referred to as inwardly rectifying K+ channels, and they have a greater tendency to allow potassium to flow into the cell rather than out of it. This asymmetry in potassium ion conductance plays a key role in the excitability of muscle cells and neurons. The protein encoded by this gene is an integral membrane protein and member of the inward rectifier potassium channel family. The encoded protein has a small unitary conductance compared to other members of this protein family. Two transcript variants encoding the same protein have been found for this gene.

1、微弱的條帶（weak single）

可能存在的原因：蛋白與抗體結(jié)合率低

解決方法:較少洗膜次數(shù)或者縮短洗膜時(shí)間；

降低洗膜液中NaCl的濃度（0.15M-0.5M）；

降低抗體稀釋液中NaCl的濃度（0.15M-0.5M）

可能存在的原因：抗體量不足

解決方法:抗體和蛋白的結(jié)合較低，可將抗體濃度調(diào)整至初始濃度的2-4倍

可能存在的原因：蛋白量不足

解決方法:增加蛋白上樣量

可能存在的原因：酶和底物時(shí)效

解決方法:將酶和底物進(jìn)行反應(yīng)，觀察現(xiàn)象，出現(xiàn)異常，則需要重新標(biāo)記或者購(gòu)買

可能存在的原因：過(guò)期或者陳舊的ECL發(fā)光液

解決方法:更換的新的ECL發(fā)光液

可能存在的原因：脫脂奶粉可能會(huì)掩蓋一些抗原

解決方法:降低牛奶的百分比或者用BSA代替

2、額外的條帶（Extra Bands）

可能存在的原因：一抗的非特性結(jié)合

解決方法:降低抗體濃度；減少總蛋白的上樣量；免疫親和層析純化抗體

可能存在的原因：二抗的非特異性結(jié)合

解決方法:跑一個(gè)二抗的陰性對(duì)照（不孵育一抗）；將二抗進(jìn)行免疫親和層析

可能存在的原因：一抗或者二抗的非異性結(jié)合

解決方法:添加0.1%-0.5%的吐溫20到抗體稀釋也中；用2%的脫脂奶粉溶解抗體，并適當(dāng)調(diào)整抗體濃度；增加洗膜液中NaCl的濃度（0.15M-0.5M）；增加洗膜時(shí)間或者次數(shù)；

可能存在的原因：蛋白聚急

解決方法:提高DTT的濃度，加樣前加熱煮沸5-10min

可能存在的原因：蛋白的降解

解決方法:避免反復(fù)凍融；添加蛋白酶抑制劑；制備新鮮樣品；

可能存在的原因：試劑污染

解決方法:配置新鮮的給中溶液
