保存條件: Store at -20 °C for one year. Avoid repeated freeze/thaw cycles. The lyophilized antibody is stable at room temperature for at least one month and for greater than a year when kept at -20°C. When reconstituted in sterile pH 7.4 0.01M PBS or diluent of antibody the antibody is stable for at least two weeks at 2-4 °C.

Anti-Gli2抗體,轉(zhuǎn)錄因子Gli2抗體產(chǎn)品介紹：It has long been known that the overexpression of either Wnt-1 or the GLI proteins results in cancer; however, the molecular basis for this transformation was poorly understood. The Wnt-1 and GLI proteins have now been placed in a signaling cascade downstream of the mammalian homologs of the Drosophila hedgehog and patched proteins. The Drosophila segment polarity gene hedgehog (hh) encodes a secreted protein that appears to function in embryonic and imaginal disc patterning. The ptc gene, also identified as a Drosophila segment polarity gene, encodes the transmembrane protein patched, the expression of which is precisely regulated during embryonic development. Hedgehog has been shown to enhance the expression of the Wnt family of proteins through a signaling cascade involving the GLI transcription factors, while patched functions as a repressor opposing the effects of hedgehog. Mutations in the ptc gene, which result in unregulated hedgehog signaling, have been correlated with the most common type of cancer, basal cell carcinoma, which affects 750,000 individuals annually in the United States alone.

合適的抗體稀釋度：

抗體的濃度是免疫染色的關(guān)鍵，如果抗體濃度過高，抗體分子過多于抗原決定簇，可導(dǎo)致抗體結(jié)合減少，產(chǎn)生陰性結(jié)果。此陰性結(jié)果并不一定是缺少抗原，而是由于抗體過量，這種現(xiàn)象類似于凝集反應(yīng)中的前帶效應(yīng)（prozone effect）。因此，必須使用一系列稀釋做“棋盤式效價滴定”，檢測抗體的合適稀釋度，以得到zui大強度的特異性染色和zui弱的背景染色。抗體稀釋度應(yīng)根據(jù)：1.抗體效價高，溶液中特異性抗體濃度越高，工作稀釋度越高；2.一般講，應(yīng)用的抗體稀釋度越大，溫育時間越長；3.對于抗體與非特異性蛋白的結(jié)合，只有高稀釋度時才能防止其非特異性背景染色；4.稀釋用緩沖液的種類，標(biāo)本的固定和處理過程等也可影響稀釋度，所以合適的稀釋度應(yīng)根據(jù)具體情況測定。抗體的稀釋主要是指*抗體，因為*抗體中特異性抗體合適的濃度是關(guān)鍵，應(yīng)用高稀釋度*抗體僅高親和力的特異性染色反應(yīng)，減少或消除其中交叉抗體反應(yīng)。

抗原修復(fù)方法：
方法1：沸水浴修復(fù)，將盛有修復(fù)液和玻片的燒杯置于沸水浴環(huán)境，保持外部沸騰狀態(tài)15min，自然冷卻至室溫。
方法2：微波修復(fù)，將盛有修復(fù)液和玻片的燒杯置于微波爐中，高火5min，停火3min，中火5min，自然冷卻至室溫。
方法3：高壓修復(fù)，修復(fù)液加入高壓鍋加熱至沸騰，放入玻片，封蓋加壓持續(xù)加熱至噴氣時開始計時修復(fù)2min，自然冷卻至室溫。
方法4：使用*修復(fù)，將*修復(fù)液加入到組織上，37℃，消化30min
