保存條件:Store at -20 °C for one year. Avoid repeated freeze/thaw cycles. The lyophilized antibody is stable at room temperature for at least one month and for greater than a year when kept at -20°C. When reconstituted in sterile pH 7.4 0.01M PBS or diluent of antibody the antibody is stable for at least two weeks at 2-4 °C.

英文介紹:This reference sequence was derived from multiple replicate ESTs and validated by similar human genomic sequence. This gene encodes a member of a family of small membrane proteins that share a 35-amino acid signature sequence domain, beginning with the sequence PFXYD and containing 7 invariant and 6 highly conserved amino acids. The approved human gene nomenclature for the family is FXYD-domain containing ion transport regulator. Transmembrane topology has been established for two family members (FXYD1 and FXYD2), with the N-terminus extracellular and the C-terminus on the cytoplasmic side of the membrane. FXYD2, also known as the gamma subunit of the Na,K-ATPase, regulates the properties of that enzyme. FXYD1 (phospholemman), FXYD2 (gamma), FXYD3 (MAT-8), FXYD4 (CHIF), and FXYD5 (RIC) have been shown to induce channel activity in experimental expression systems. This gene product, FXYD7, is novel and has not been characterized as a protein.

Anti-FXYD7抗體,FXYD離子轉運調節因子7抗體轉膜(Transfer)
我們*在Western實驗中選用PVDF膜。硝酸纖維素膜(NC膜)或PVDF膜(二氟化樹脂膜膜孔徑:0.45um)也可以使用,但硝酸纖維素膜比較脆,在操作過程中特別是用鑷子夾取等過程中容易裂開。膜的使用請參考生產商的*使用步驟。
通常如果使用Bio-Rad的標準濕式轉膜裝置,可以設定轉膜電壓120V,轉膜時間為30-60分鐘。具體的轉膜時間要根據目的蛋白的大小而定,目的蛋白的分子量越大,需要的轉膜時間越長,目的蛋白的分子量越小,需要的轉膜時間越短。
在轉膜過程中,特別是高電流快速轉膜時,通常會有非常嚴重的發熱現象,把轉膜槽放置在冰浴中進行轉膜。轉膜的效果可以觀察所使用的預染蛋白質分子量標準,通常分子量zui大的1-2條帶較難全部轉到膜上。轉膜的效果也可以用麗春紅染色液對膜進行染色,以觀察實際的轉膜效果。也可以用考馬斯亮藍快速染色液對完成轉膜的SDS-PAGE膠進行染色,以觀察蛋白的殘留情況。
