一抗孵育(Primary antibody incubation)
參考一抗的說明書，按照適當比例用Western一抗稀釋液稀釋一抗。
用微型臺式真空泵吸盡封閉液，立即加入稀釋好的一抗，室溫或4℃在側擺搖床上緩慢搖動孵育一小時。如果一抗孵育一小時效果不佳，可以在4℃緩慢搖動孵育過夜。回收一抗。加入Western洗滌液，在側擺搖床上緩慢搖動洗滌5-10分鐘。吸盡洗滌液后，再加入洗滌液，洗滌5-10分鐘。共洗滌3次。如果結果背景較高可以適當延長洗滌時間并增加洗滌次數。
二抗孵育(Secondary antibody inucubation)
參考二抗的說明書，按照適當比例用Western二抗稀釋液稀釋辣根過氧化物酶(HRP)標記的二抗。
用微型臺式真空泵吸盡洗滌液，立即加入稀釋好的二抗，室溫或4℃在側擺搖床上緩慢搖動孵育一小時。回收二抗，加入Western洗滌液，在側擺搖床上緩慢搖動洗滌5-10分鐘。吸盡洗滌液后，再加入洗滌液，洗滌5-10分鐘。共洗滌3次。如果結果背景較高可以適當延長洗滌時間并增加洗滌次數。

英文介紹：Chromosome 1 is the largest human chromosome spanning about 260 million base pairs and making up 8% of the human genome. There are about 3,000 genes on chromosome 1, and considering the great number of genes there are also a large number of diseases associated with chromosome 1. Notably, the rare aging disease Hutchinson-Gilford progeria is associated with the LMNA gene which encodes lamin A. When defective, the LMNA gene product can build up in the nucleus and cause characteristic nuclear blebs. The mechanism of rapidly enhanced aging is unclear and is a topic of continuing exploration. The MUTYH gene is located on chromosome 1 and is partially responsible for familial adenomatous polyposis. Stickler syndrome, Parkinsons, Gaucher disease and Usher syndrome are also associated with chromosome 1. A breakpoint has been identified in 1q which disrupts the DISC1 gene and is linked to schizophrenia. Aberrations in chromosome 1 are found in a variety of cancers including head and neck cancer, malignant melanoma and multiple myeloma. The C1orf109 gene product has been provisionally designated C1orf109 pending further characterization.
