轉膜(Transfer)
我們*在Western實驗中選用PVDF膜。硝酸纖維素膜(NC膜)或PVDF膜（二氟化樹脂膜膜孔徑：0.45um)也可以使用，但硝酸纖維素膜比較脆，在操作過程中特別是用鑷子夾取等過程中容易裂開。膜的使用請參考生產商的*使用步驟。
通常如果使用Bio-Rad的標準濕式轉膜裝置，可以設定轉膜電壓120V，轉膜時間為30-60分鐘。具體的轉膜時間要根據目的蛋白的大小而定，目的蛋白的分子量越大，需要的轉膜時間越長，目的蛋白的分子量越小，需要的轉膜時間越短。
在轉膜過程中，特別是高電流快速轉膜時，通常會有非常嚴重的發熱現象，把轉膜槽放置在冰浴中進行轉膜。轉膜的效果可以觀察所使用的預染蛋白質分子量標準，通常分子量zui大的1-2條帶較難全部轉到膜上。轉膜的效果也可以用麗春紅染色液對膜進行染色，以觀察實際的轉膜效果。也可以用考馬斯亮藍快速染色液對完成轉膜的SDS-PAGE膠進行染色，以觀察蛋白的殘留情況。

封閉(Blocking)
轉膜完畢后，立即把蛋白膜放置到預先準備好的Western洗滌液中，漂洗1-2分鐘，以洗去膜上的轉膜液。從轉膜完畢后所有的步驟，一定要注意膜的保濕，避免膜的干燥，否則極易產生較高的背景。
用微型臺式真空泵吸盡洗滌液，加入Western封閉液，在搖床上緩慢搖動,室溫封閉60分鐘。對于一些背景較高的抗體，可以在4℃ 封閉過夜。

英文介紹：c-Kit is a transmembrane tyrosine kinase encoded by the cKit proto oncogene. c-Kit acts to regulate a variety of biological responses including cell proliferation, apoptosis, chemotaxis and adhesion. Ligand binding to the extracellular domain leads to autophosphorylation on several tyrosine residues within the cytoplasmic domain, and activation. Mutations in c-Kit have been found to be important for tumor growth and progression in a variety of cancers including mast cell diseases, gastrointestinal stromal tumor, acute myeloid leukemia, Ewing sarcoma and lung cancer. Phosphorylation at tyrosine 721 of c-Kit allows binding and activation of PI3 kinase.

保存條件：Store at -20 °C for one year. Avoid repeated freeze/thaw cycles. The lyophilized antibody is stable at room temperature for at least one month and for greater than a year when kept at -20°C. When reconstituted in sterile pH 7.4 0.01M PBS or diluent of antibody the antibody is stable for at least two weeks at 2-4 °C.
