英文介紹：First discovered in C. elegans, Hemicentins are extracellular matrix (ECM) proteins that contain a single von Willebrand A domain at the amino terminus, more than 40 tandem immunoglobulin domains, multiple tandem epidermal growth factors, and a single fibulin-like carboxy-terminal module. In mammals, Hemicentins are expressed in the ECM of eye, skin and tongue epithelium and in the ECM of some blood vessels. Due to the expression pattern in areas that are subject to a significant amount of stress, it is thought that Hemicentins likely play a role in the architecture of flexible and adhesive cell junctions. Hemicentin-2 is a 5,065 amino acid secreted protein that is expressed in the extracellular matrix. There are three isoforms of Hemicentin-2 that are produced as a result of alternative splicing events

保存條件：Store at -20 °C for one year. Avoid repeated freeze/thaw cycles. The lyophilized antibody is stable at room temperature for at least one month and for greater than a year when kept at -20°C. When reconstituted in sterile pH 7.4 0.01M PBS or diluent of antibody the antibody is stable for at least two weeks at 2-4 °C.

1 脫蠟和水化：脫蠟前應(yīng)將組織芯片在室溫中放置60分鐘或60℃恒溫箱中烘烤20分鐘。 a 組織芯片置于二甲苯中浸泡10分鐘,更換二甲苯后在浸泡10分鐘
b 無水乙醇中浸泡五分鐘
c 95%乙醇中浸泡五分鐘
d 75%乙醇中浸泡五分鐘
2 抗原修復(fù)：用于福爾馬林固定的石蠟包埋組織芯片：
A 抗原熱修復(fù)
a 高壓熱修復(fù) 在沸水中加入EDTA（ph8.0）或0.01m*緩沖溶液（ph6.0）.蓋上不銹鋼鍋蓋，但不能鎖定。將玻片置于金屬染色加上，緩慢加壓，是玻片在緩沖液中浸泡五分鐘，然后將蓋子鎖定，小閥門將會(huì)升起來。10分鐘后除去熱源，置入涼水中，當(dāng)小閥門沉下去后打開蓋子。此方法適用于較難檢測(cè)或核抗原的抗原修復(fù)。
b 沸熱修復(fù) 電爐或水浴鍋加熱0.01*緩沖液（ph6.0）至95℃左右，放入組織芯片加熱10-15分鐘。
C 微波爐加熱 在微波爐里加熱0.01*緩沖液（ph6.0）至沸騰后將組織芯片放入，斷電，間隔5-10分鐘，反復(fù)1-2次。適用的抗原：
Bcl-2.Bax.AR.PR.C-fos.x-jum.z-kit.c-myc.E-cadherin.
ChromograninA.Cyclin.ER.Heatshock.Protein.HPV.Ki-67.MDMZ.P53.P34.P15.P-glycoprotein.PKC.PCNA.ras.Rb.等
B 酶消化方法 
常用0.1%*和0.4%*液。*使用前預(yù)熱37℃，消化時(shí)間約為5-30分鐘。適用于被固定遮蔽的抗原：包括 Collagen.GFAP.
Complement.Cytokeratin.C-erB-2.LCA.LN.等
