【Anti-TIMP-2抗體】金屬蛋白酶組織抑制因子2抗體產品介紹：This gene is a member of the TIMP gene family. The proteins encoded by this gene family are natural inhibitors of the matrix metalloproteinases, a group of peptidases involved in degradation of the extracellular matrix. In addition to an inhibitory role against metalloproteinases, the encoded protein has a unique role among TIMP family members in its ability to directly suppress the proliferation of endothelial cells. As a result, the encoded protein may be critical to the maintenance of tissue homeostasis by suppressing the proliferation of quiescent tissues in response to angiogenic factors, and by inhibiting protease activity in tissues undergoing remodelling of the extracellular matrix.
保存條件: Store at -20℃ for one year. Avoid repeated freeze/thaw cycles. The lyophilized antibody is stable at room temperature for at least one month and for greater than a year when kept at -20℃. When reconstituted in sterile pH 7.4 0.01M PBS or diluent of antibody, the antibody is stable for at least six weeks at 2-4 ℃
抗原修復（免疫組化石蠟切片）
用于福爾馬林固定的石蠟包埋組織芯片需要抗原修復。
福爾馬林固定的組織有可能破壞了原來組織的抗原結構，蛋白多肽發生交聯，導致部分抗原表位封閉，結合位點減少，所以需要抗原修復，以暴露原來的抗原表位，達到充分顯露抗原，以不出現明顯脫片現象為佳。
抗原修復的幾種常用方法（供參考）
1、 微波爐加熱：
在微波爐里加熱0.01*緩沖液（pH6.0）至沸騰后將組織芯片放入，斷電，間隔5-10分鐘，反復1-2次。根據玻片情況可參考采用微波加熱“3-5-3法"3分鐘溫火沸騰后靜止5分鐘，再溫火沸騰3分鐘即可。
適用的抗原：來源于人、大鼠、小鼠的組織標本；來源于其他動物種屬的組織標本。
2、 沸熱修復：
電爐或水浴鍋加熱0.01M*緩沖液（pH6.0）至95℃左右，放入組織芯片加熱10-15分鐘。
3、 高壓熱修復：
在沸水中加入EDTA（pH8.0）或0.01m*緩沖溶液（pH6.0）.蓋上不銹鋼鍋蓋，但不能鎖定。將玻片置于金屬染色架上，緩慢加壓，使玻片在緩沖液中浸泡五分鐘，然后將蓋子鎖定，小閥門將會升起來。10分鐘后除去熱源，置入涼水中，當小閥門沉下去后打開蓋子。此方法適用于較難檢測或核抗原的抗原修復。（骨及軟骨組織的標本選擇較溫和的微波加熱修復） 。
4、 酶消化方法：
常用0.1%*和0.4%*液。*使用前預熱37℃，消化時間約為5-30分鐘。
適用于被固定遮蔽的抗原：包括 Collagen.GFAP. Complement.Cytokeratin.c-erB-2.LCA.LN.等。
